Quantitation of imidazoles by agar-disk diffusion.
A simple, reliable, and inexpensive assay for quantitation of the imidazole drugs miconazole, RV 40,500, and RV 41,400 was tested. The assay, similar to a Kirby-Bauer test, was sensitive to less than or equal to 0.2 mug of drug per ml and linear from less than or equal to 0.2 to 10.0 mug/ml. Concentration of inoculum and agar depth in test plates was not as critical as the type of medium, amoun...
متن کاملMezlocillin susceptibility tests by the single-disk agar diffusion method.
A regression line for correlating zones of inhibition of a 75-mug mezlocillin disk with minimal inhibitory concentrations of mezlocillin for Enterobacteriaceae and nonfermenters was calculated. Correlation was excellent (r = -0.96) for all species tested except Pseudomonas cepacia.
متن کاملEvaluation of Mueller-Hinton agar for disk diffusion susceptibility tests.
Twelve lots of commercially prepared Mueller-Hinton agar from four medium manufacturers were evaluated for performance with recommended quality control organisms, medium pH, agar depth and uniformity, and characteristics of the zones of inhibition. Only 2 of 12 lots were acceptable for disk diffusion susceptibility tests. Significant problems were observed with the preparation and pouring of th...
متن کاملAnalysis of rifampin disk diffusion and stability in 7H10 agar.
Rifampin was incorporated into Middlebrook 7H10 medium either by adding an aliquot of the antibiotic into melted agar (final concentration 1.0 and 3.0 mug/ml) or by submerging a 5 or 15 mug of rifampin paper disk into 5 ml of melted agar contained in one quadrant of a Felson "X" plate. At intervals, plugs of agar were removed from the stored plates and assayed. Plates stored at 5 C for 28 days ...
متن کاملCorrelation of netilmicin agar dilution and disk diffusion susceptibilities.
A study of 283 isolates of gram-negative bacilli revealed a good correlation (r = -0.74) between disk diffusion zones of inhibition and agar dilution minimal inhibitory concentrations. Regression analysis suggested that strains with zone sizes =/<11 mm should be considered resistant, but 34 of 45 strains resistant by minimal inhibitory concentration (including 27 strains of Pseudomonas aerugino...
